Ashwagandha KSM-66: why the extract matters as much as the plant

A
Adam DALI

Two jars of ashwagandha, two very different labels… and yet the same plant on the front. So, where is the difference? It hides in a word that many consumers gloss over: extract.

At Pure Lab Nutrition, we have chosen the KSM-66® extract for our ORGANIC Ashwagandha. In this article, we explain why the form of the plant matters just as much as the plant itself, how to read a label without getting trapped, and which mistakes to avoid when choosing.

Powder or extract: what is the difference?

🌾 Root powder

Powder is simply the dried and ground root. Nothing is concentrated: the content of active compounds depends entirely on the harvested plant (soil, climate, age of the root). To obtain quantities comparable to those in studies, one often needs to take several grams per day.

🧪 The extract

The extract is obtained by isolating and concentrating the plant's compounds of interest from the root. The result: a smaller dose that is easier to take, and above all, a better-controlled composition. Furthermore, it is in the form of root extracts that ashwagandha has been tested in most recent clinical trials.

📋 Comparison at a glance

Root powder Standardized extract
Concentration That of the raw root Concentrated compounds of interest
Active content Variable depending on the harvest Indicated by the titration
Quantity to take Often several grams A few hundred milligrams
Link to studies Difficult to compare Comparable if the extract is identified

Standardization, a guarantee of consistency

A "standardized" extract guarantees a precise content of active compounds from one batch to the next. For ashwagandha, we generally measure withanolides, the most studied compounds in the plant.

✅ Why it is important

  • Consistency: you take the same amount of active ingredients every day, from one jar to another
  • Comparability: you can compare your dose to those used in studies
  • Transparency: a brand that states its titration is committed to what it sells

Key takeaway: a higher percentage is not automatically better. What counts is a root extract, with a clearly indicated content, and whose effectiveness has been evaluated.

🧮 A practical case: decoding a titration

Let’s take a capsule containing 300 mg of extract standardized to 5% withanolides. The calculation is simple: 5% of 300 mg equals 15 mg of withanolides per capsule. If another label only says "500 mg of ashwagandha" without specifying whether it is powder or extract, or its titration, you simply cannot know what you are taking. This is the whole point of standardization: it makes products comparable.

KSM-66®: what are we talking about?

🏷️ A branded extract

KSM-66® is an ashwagandha extract developed by the Indian company Ixoreal Biomed. According to its manufacturer, it is obtained solely from roots, via a chemical solvent-free extraction process, and standardized to 5% withanolides.

🔬 An extract used in clinical trials

Its main advantage is that it has been used in randomized, placebo-controlled trials published in scientific journals, for example:

  • Stress: a trial on 64 stressed adults, published in 2012 in the Indian Journal of Psychological Medicine (Chandrasekhar et al.), with 600 mg per day for 60 days; cortisol levels decreased more than with a placebo
  • Sleep: a trial on 60 adults suffering from insomnia, published in 2019 in Cureus (Langade et al.), with 600 mg per day for 10 weeks

For the details of the results on sleep, read our article Ashwagandha and sleep: what the studies really say.

Full transparency: some of these studies were funded or supported by the extract's manufacturer, which is common in this field. They remain useful, but deserve to be confirmed by independent research. We also remind you that no health claims for ashwagandha are authorized to date in the European Union: we are talking about study results, not guaranteed effects.

🔁 Other studied extracts

KSM-66® is not the only extract to have been tested. In 2019, a trial published in the journal Medicine evaluated another ashwagandha extract at a dose of 240 mg per day in 60 stressed adults for 60 days. Anxiety scores and morning cortisol levels decreased more than with a placebo. This example perfectly illustrates our point: each extract has its own composition and studied dose, and this data cannot be transposed from one product to another.

⚖️ What about safety?

A high-quality extract does not exempt you from precautions. In 2024, the French agency ANSES advised against ashwagandha, regardless of its form, for pregnant or breastfeeding women, minors under 18, people with thyroid, liver, or heart disorders, and those taking sedative medications.

How to read an ashwagandha label

What to look for Why
"Root extract" The traditional and most studied part
Withanolide titration Guarantees a constant active content
Amount of extract per dose To compare with studied doses
Name of the extract (if branded) Allows you to find the studies that used it
Organic label Quality of the raw material
Warnings A serious brand lists contraindications

Common mistakes

❌ Comparing milligrams that cannot be compared

500 mg of powder and 300 mg of standardized extract do not contain the same things. Before comparing two products, always check whether it is powder or extract, and look at the titration.

❌ Believing that "more concentrated" means "better"

A very high titration has not necessarily been studied. It is better to have an extract whose composition matches that used in published trials.

❌ Trying to replicate study doses on your own

The trials used precise protocols under supervision. Follow the dose indicated on your supplement label and seek advice from a healthcare professional if you are considering doing otherwise.

❌ Forgetting contraindications

The best extract in the world is still not recommended for the groups mentioned by ANSES. If you are currently taking medication, speak to your doctor first.

Frequently asked questions

❓ Is KSM-66® the only good extract?

No. Other extracts exist, and some have also been studied. The advantage of KSM-66® is that it is an identified root extract with a known titration, which makes it possible to locate the published studies that used it.

❓ Why only the root?

It is the part used traditionally and the one that is the subject of the majority of recent clinical trials. A product that mixes root and leaves is not comparable to studies conducted on the root alone.

❓ What does "standardized to 5%" mean?

It means that 5% of the extract's mass consists of withanolides. For 300 mg of extract, that represents 15 mg of withanolides.

In summary

1️⃣ Extract rather than powder: a better-controlled composition at a lower dose
2️⃣ Root and titration: the two items to look for first
3️⃣ A studied extract: KSM-66® has been used in several clinical trials
4️⃣ The precautions remain the same, regardless of the extract
